Ultra-Fast Quantitative Analysis of Drugs in Plasma by LC-MS/MS
In the early stages of drug discovery DMPK analysis, large-scale high-throughput HPLC plays a crucial role, and ultra-fast HPLC columns are essential.
The combination of the high-pressure specification 3μm particle high-throughput column Cadenza CD-C18 HT (50x2mm), Shimadzu UFLC, and AB Sciex API5000 enabled analysis in as fast as 9.6 seconds. Additionally, it achieved equivalent peak capacity at 1/4 the pressure of sub-2μm particles and 60% of core-shell particles.
In ultra-fast analysis using small columns, it is believed that the influence of extra-column diffusion is greater than particle size. The low-pressure property of the 3μm particle column is advantageous at high flow rates regarding pressure limits.
The Cadenza CD-C18 HT (high-pressure specification), which balances column pressure and separation performance, is actively utilized in DMPK analysis.
